The Cell Sorts CS-0037 Mouse Pan B Cell Sorting Kit uses column-free negative selection to isolate untouched Pan B cells from mouse spleen, lymph nodes, bone marrow, and peritoneal lavage with up to 98.8% purity in ~25 min. Grind tissue through a 70 μm cell strainer and perform RBC lysis with ACK buffer. 2 μL Biotin-Antibody Mix labels non-B cells at 4°C for 10 min. After wash/centrifugation (500×g, 5 min), 20 μL Beads Streptavidin bind at 4°C for 10 min. Magnetic stand 5 min, then pour off purified Pan B cells. A final 500×g spin concentrates cells for downstream use.

Furthermore, this streamlined protocol ensures consistent results across experiments.
How the Mouse Pan B Cell Sorting Kit Works
Simple 4-step protocol for rapid cell isolation
Step 1: Sample Preparation
Grind mouse spleen, lymph node, or bone marrow through a 70 μm cell strainer, perform red blood cell lysis with ACK buffer, wash, filter again, count cells, and resuspend in separation buffer at 1×10⁸ cells/mL.
Step 2: Antibody Labeling
Aliquot 100 μL cell suspension (1×10⁷ cells), add 2 μL Biotin-Antibody Mix, mix well, incubate at 4°C for 10 min. Wash with 1 mL separation buffer, centrifuge at 500×g for 5 min, resuspend in 100 μL separation buffer.
Step 3: Magnetic Bead Binding
Add 20 μL pre-washed Beads Streptavidin, mix well, incubate at 4°C for 10 min to bind labeled non-B cells.
Step 4: Column-Free Magnetic Separation & Collection
Dilute with 2.5 mL separation buffer, mix thoroughly by pipetting 5 times, place on magnetic stand for 5 min. Gently pour supernatant into sterile centrifuge tube while keeping tube on magnet. Centrifuge at 500×g for 5 min and resuspend for downstream use.
