Human T Cell Sorting Kit

Column-Free Negative Human

The Human T Cell Sorting Kit (CS-0014) uses column-free negative selection to isolate untouched pan T cells (CD4+ and CD8+) from human PBMCs with 90–99% purity in ~15 min. Biotin-conjugated antibody cocktails label non-target cells (B cells, monocytes, NK cells, granulocytes), and streptavidin nanobeads bind them for magnetic removal — leaving untouched, antibody-free T cells in their native, unactivated state. The entire pan T cell sorting protocol requires no columns, no centrifugation, and no contact between beads and target cells.

Compared to density gradient centrifugation (40–60% purity) or FACS sorting (expensive, low throughput, shear stress), CS-0014 delivers 90–99% T cell purity with >95% recovery and >95% viability in just ~15 minutes. Because no beads or antibodies contact target cells, isolated T cells fully retain their TCR complex and CD3 signaling capacity — making this kit ideal for CAR-T development, TCR-T engineering, comprehensive T cell studies, and adoptive cell therapy research.

~15 min fast sorting — Rapid protocol: 5 min antibody incubation + 5 min bead binding + 3 min magnetic separation
High viability — Gentle isolation preserves TCR complex, CD3 signaling, and full functional activity for downstream assays
Ready for downstream use — Compatible with flow cytometry, cell culture, CAR-T development, TCR-T engineering, and adoptive cell therapy

The Human T Cell Sorting Kit uses column-free negative selection to isolate untouched Pan T Cells from human PBMCs. To achieve these results, follow this simple 4-step protocol that takes just ~30 min from start to finish. No centrifugation and no lengthy incubations. Furthermore, this streamlined protocol ensures consistent results across experiments.

How the Human T Cell Sorting Kit Works

Simple 4-step protocol for rapid cell isolation

1
Step 1 of T cells sorting: Sample Preparation - Prepare PBMC single-cell suspension with FcR Blocker for Human T Cell Sorting Kit CS-0014

Step 1: Sample Preparation

Prepare a PBMC single-cell suspension, wash cells, and resuspend in separation buffer at 5×10⁶ cells/mL (e.g., 1×10⁷ cells in 5 mL).

2
Step 2 of T cells sorting: Antibody Cocktail Labeling - Biotin-conjugated antibodies target non-T cells for depletion with CS-0014

Step 2: Antibody Labeling

Add T cell negative selection cocktail (10 μL per 1×10⁷ cells), mix well, and incubate at room temperature for 5 min. Antibodies label non-T cells (B cells, monocytes, NK cells, granulocytes).

3
Step 3 of T cells sorting: Magnetic Nanobead Binding - Streptavidin nanobeads bind labeled non-target cells for magnetic removal with CS-0014

Step 3: Magnetic Bead Binding

Add magnetic nanobeads (8 μL per 1×10⁷ cells), mix well, and incubate at room temperature for 5 min. Nanobeads bind labeled non-target cells for magnetic removal.

4
Step 4 of T cells sorting: Column-Free Separation - Purified T cells collected by magnetic separator pour-off for CS-0014

Step 4: Column-Free Separation & Collection

Place tube on magnetic separator for 3 min. Pour off supernatant containing purified, untouched pan T cells. Ready for CAR-T development, flow cytometry, or adoptive cell therapy.

In addition, the Human T Cell Sorting Kit is compatible with a wide range of downstream applications, making it a versatile tool for your immunology research workflow.

Human T Cell Sorting Kit — Frequently Asked Questions

The CS-0014 Human T Cell Sorting Kit uses column-free negative selection. Non-target cells (B cells, monocytes, NK cells, granulocytes) are labeled with a biotin-conjugated antibody cocktail and removed by magnetic separation, while untouched T cells remain in the supernatant. This ensures no beads or antibodies ever contact the target cells, preserving their native functional state.
Purity of isolated T cells typically reaches 90-99%, as validated by flow cytometry. The optimized antibody cocktail efficiently depletes all non-target cell populations while maximizing recovery of viable, functional T cells from human PBMCs.
The entire column-free protocol takes approximately 18-20 minutes from start to finish: 5-10 min for antibody cocktail incubation, 5-10 min for magnetic nanobead binding, and 2-3 min for magnetic separation. No column washing or centrifugation steps are required.
Antibodies and streptavidin nanobeads bind exclusively to non-target cells—they never touch T cells. This untouched approach ensures full preservation of surface receptors and native functional states, making the isolated cells ideal for downstream functional assays and cell therapy research.
Step 1: Prepare a PBMC single-cell suspension and add Human FcR Blocker.
Step 2: Incubate with biotin-conjugated antibody cocktail targeting non-target cells at 4°C for 5-10 min.
Step 3: Add streptavidin nanobeads and incubate for 5-10 min.
Step 4: Place in magnetic separator for 2-3 min, then pour off untouched T cells.
Total time: 18-20 minutes.
Yes, the CS-0014 kit is ideal for CAR-T cell therapy research. The untouched, bead-free pan T cells (both CD4+ and CD8+) maintain full TCR functionality and are immediately ready for lentiviral transduction with CAR constructs. The high purity (90–99%) and preserved activation state ensure optimal transduction efficiency and therapeutic potency.
The antibody cocktail targets CD14, CD16, CD19, CD36, CD56 on non-target cells—effectively labeling B cells, monocytes, NK cells, granulocytes for depletion while leaving T cells completely untouched. This comprehensive marker panel ensures high purity and recovery of the target cell population.
Isolated T cells are immediately ready for: flow cytometry and cell surface marker analysis, cell culture and expansion, functional assays, CRISPR gene editing, single-cell RNA sequencing (scRNA-seq), cell receptor sequencing, and cell-targeted drug screening. Zero bead or antibody interference means no additional washing steps are required.
We recommend flow cytometry using appropriate antibody staining. Typical results show 90-99% target cells in the collected fraction. Viability can be assessed using Trypan blue exclusion or 7-AAD staining. For best results, gate on lymphocytes by forward and side scatter, then analyze marker expression within the gated population.
The CS-0014 uses a broader depletion cocktail targeting all non-T cells to isolate pan T cells. The CS-0003 specifically targets CD3+ T cells. Both use column-free negative selection with 90–99% purity in 18–20 min. The CS-0014 is optimized for comprehensive T cell isolation including rare subsets; the CS-0003 is our standard CD3+ T cell kit.
